Les travaux dans RÉALISE de 2007 à 2014

Nous étudions les possibilités d’utiliser des sidérophores bactériens (pyoverdine, pyocheline et azotobactine) ou des analogues de synthèse pour la détection et la quantification de métaux toxiques à l’état de traces.

Nous étudions également les possibilités d’utiliser les sidérophores bactériens et les voies d’assimilation du fer des bactéries dans des procédés de bioremédiation. Nous avons déjà montré que le sidérophore pyoverdine pouvait chélater de nombreux métaux toxiques et que ces complexes semblent interagir avec les récepteur protéiques au niveau de la membrane externe de Pseudomonas aeruginosa. Nous allons déterminer pour chacun de ces complexes ses possibilités d’être transporté puis établir son devenir à l’intérieur de la cellule. Cette étude permettra d’évaluer le potentiel de la voie pyoverdine dans des procédés de bioremédiation. Le même type d’étude est mené avec d’autres sidérophores bactériens.

90 % des microorganismes vivent dans la nature sous forme de biofilm. De ce fait, nous étudierons également le devenir de ces complexes sidérophores-métaux toxiques en présence de Pseudomonas aeruginosa cultivé en biofilm ou en billes d’alginates (collaboration avec le Pr. Thierry Lebeau). En effet, le comportement des microorganismes selon leur mode de culture est complètement différent.

En fonction des résultats obtenus avec ces premières études et selon les métaux, nous étudierons en collaboration avec le groupe du Prof Thierry Lebeau les possibilités d’utiliser ces voies de transport de fer utilisant des sidérophores dans différents procédés de détoxification :

  • bioremédiation
  • biaugmentation
    En effet, la présence de ces bactéries productrices de sidérophores dans le sol va permettre d’augmenter la solubilité des métaux et leur biodisponibilité.
  • couplage bioaugmentation -phytoextraction
    Il s'agit dans ce cas, non pas d'immobiliser le métal mais de dépolluer le sol grâce à la phytoextraction "accélérée" par bioaugmentation de sols contaminés, suivie du traitement de la biomasse végétale par incinération. Ce projet est financé par une ANR "Jeunes chercheurs  et jeunes chercheuses" en 2005 (Réf. : ANR-05-JCJC-0181-01
